You can use the Zip 100MB disks in the Zip 250MB drive.

You cannot use the Zip^® 250MB disks with the Zip 100MB drive. The Zip 100MB drives were not designed to be forward compatible with the Zip 250MB disks. If a Zip 250MB disk is inserted into a Zip 100MB drive, the disk will automatically eject.
